The iNTS-GMMA vaccine is a bivalent biologic vaccine candidate developed by the GSK Vaccines Institute for Global Health (GVGH) for the prevention of invasive nontyphoidal Salmonella (iNTS) infections. The vaccine utilizes Generalized Modules for Membrane Antigens (GMMA) technology, which consists of outer membrane vesicles (OMVs) shed from genetically modified strains of *Salmonella enterica* serovars Typhimurium and Enteritidis. These vesicles naturally present multiple immunodominant antigens, including the O-antigen (OAg) component of bacterial lipopolysaccharide and various outer membrane proteins, to the immune system. This approach is designed to elicit a broad and protective antibody response against the two serovars most commonly associated with iNTS disease, particularly in high-burden regions like sub-Saharan Africa. The vaccine is currently being evaluated in Phase I/II clinical trials in both European and African populations, including infants, children, and adults. GVGH is also exploring a trivalent formulation (iNTS-TCV) that combines this bivalent vaccine with a typhoid conjugate vaccine.
