A combination therapy consisting of **inupadenant** (a potent, selective, non-brain-penetrant small molecule antagonist of the adenosine A2A receptor) and **standard cytotoxic chemotherapy** (including agents such as carboplatin and pemetrexed, or carboplatin and paclitaxel). This regimen is under investigation for the treatment of advanced solid tumors, particularly non-small cell lung cancer (NSCLC) after progression on immunotherapy. Inupadenant works by blocking adenosine-mediated immunosuppression in the tumor microenvironment, thereby enhancing antitumor immune responses. The chemotherapy agents act through direct cytotoxic mechanisms. The combination aims to increase therapeutic efficacy by targeting tumor cell survival both through immune-mediated and direct cytotoxic pathways. The approach is experimental and not approved for any indication.
