A combination therapy of **inupadenant**, a small molecule antagonist of the adenosine A2A receptor, and **pembrolizumab**, a humanized monoclonal IgG4 antibody that targets programmed cell death protein 1 (PD-1) on T cells. Inupadenant works by blocking adenosine-mediated immunosuppression in the tumor microenvironment, thus keeping immune cells active against cancer. Pembrolizumab inhibits the PD-1 pathway, preventing tumor cells from suppressing immune responses and restoring T-cell activity. This combination is investigational and is being studied in advanced solid tumors, melanoma (especially PD-1-resistant), and castrate-resistant prostate cancer. It is not yet approved for any indication[1][3][5][9].
