IO102 + IO103 is a first-in-class, dual-antigen therapeutic cancer vaccine developed by IO Biotech. It consists of two synthetic long peptides: IO102, which targets Indoleamine 2,3-dioxygenase (IDO), and IO103, which targets Survivin (BIRC5). IDO is an enzyme that promotes an immunosuppressive tumor microenvironment by depleting tryptophan, while survivin is an anti-apoptotic protein frequently overexpressed in various cancers. The vaccine is designed to stimulate and expand endogenous T cells that specifically recognize and eliminate both tumor cells and immunosuppressive cells (such as regulatory T cells and myeloid-derived suppressor cells) that express these proteins. By modulating the tumor microenvironment and directly targeting cancer cells, IO102 + IO103 aims to enhance the efficacy of checkpoint inhibitors, particularly PD-1/PD-L1 blockers.
