IO103 + Montanide

IO103 + Montanide is a first-in-class therapeutic cancer vaccine combination developed by IO Biotech, consisting of a 19-amino acid peptide derived from the Programmed cell death 1 ligand 1 (PD-L1) protein emulsified with the water-in-oil adjuvant Montanide ISA 51. The vaccine is designed to induce a specific T-cell response against cells that overexpress PD-L1, which include both malignant tumor cells and immunosuppressive cells within the tumor microenvironment, such as regulatory T cells and myeloid-derived suppressor cells. By stimulating PD-L1-specific cytotoxic T cells, IO103 aims to directly eliminate these cells and alleviate immune suppression, thereby enhancing the overall anti-tumor immune response. It is currently being investigated in Phase 2 clinical trials for squamous cell carcinoma of the head and neck (SCCHN), often in combination with immune checkpoint inhibitors like pembrolizumab, to evaluate its potential to improve outcomes in patients with recurrent or metastatic disease.
