IOS-1002 + pembrolizumab is an investigational combination therapy for advanced solid tumors. IOS-1002 is a first-in-class, HLA-based checkpoint inhibitor developed by ImmunOs Therapeutics that targets the immune inhibitory receptors LILRB1 (ILT2), LILRB2 (ILT4), and KIR3DL1 on innate and adaptive immune cells. By blocking these receptors, IOS-1002 enhances anti-tumor immunity by promoting infiltration of M1 macrophages, T cells, and NK cells into the tumor microenvironment[5][1][2]. Pembrolizumab (Keytruda) is a humanized monoclonal antibody that inhibits PD-1 on lymphocytes to prevent cancer-mediated immune evasion[6]. The combination aims to synergistically enhance anti-tumor responses in patients with advanced solid tumors who may not respond adequately to single-agent immunotherapy. This regimen is currently being evaluated in phase 1 clinical trials as both monotherapy and combination therapy[3][4].
