Overview

- **IOV-2001 + aldesleukin** is a combination investigational therapy for hematologic malignancies, specifically chronic lymphocytic leukemia (CLL) and small lymphocytic lymphoma (SLL). - **IOV-2001** is an autologous, non-genetically modified, polyclonal T cell product composed mainly of TCRαβ+ effector memory T cells (PBLs), enriched and expanded from the patient’s own peripheral blood mononuclear cells (PBMC). It is designed to restore antitumor T-cell function in patients with T-cell exhaustion by providing cytotoxic, antigen-specific, HLA-dependent T cells[2][4]. - **Aldesleukin** is a recombinant interleukin-2 cytokine (IL-2) that acts as a T-cell growth factor and is used to stimulate the expansion, activation, and persistence of T cells, both in vivo and ex vivo. It is approved (as Proleukin) as an antineoplastic and immunotherapeutic agent, and in this combination is used to support the growth of IOV-2001 T cells before infusion and/or may be administered alongside the product[1][3]. - The combination leverages IOV-2001’s tumor-targeted cytotoxicity and aldesleukin’s immune-stimulatory role, aiming to overcome T-cell dysfunction in relapsed/refractory CLL or SLL. - IOV-2001 + aldesleukin is being developed by Iovance Biotherapeutics for use in patients with CLL or SLL refractory to prior therapies, especially after BTK-inhibitor treatment[2][4].
