Overview

**Ipatasertib + abiraterone** is an investigational drug combination composed of ipatasertib, a selective ATP-competitive inhibitor of AKT (protein kinase B), and abiraterone acetate, an androgen biosynthesis inhibitor targeting CYP17A1. This combination is being developed primarily for the treatment of metastatic castration-resistant prostate cancer (mCRPC), especially in patients with PTEN-loss or activation of the PI3K/AKT pathway. The mechanism of action targets dual blockade of androgen receptor signaling (abiraterone) and the PI3K/AKT pathway (ipatasertib), which are both implicated in prostate cancer progression and treatment resistance. The combination has shown increased progression-free survival in patients whose tumors exhibit PTEN loss or related pathway activation, and is generally administered with prednisone/prednisolone. The combination is not currently approved for clinical use.
