Overview

This combination therapy regimen consists of the AKT inhibitor ipatasertib, the androgen receptor antagonist darolutamide, and standard androgen deprivation therapy (ADT). It is being investigated in the ADDItion trial (BTCRC-GU19-404) for the treatment of prostate cancer. The regimen targets both the androgen receptor signaling pathway and the PI3K/AKT pathway, which are often co-activated in advanced prostate cancer and contribute to treatment resistance. The study, led by Dr. David VanderWeele, evaluates the safety and efficacy of this triple combination in patients with metastatic castration-resistant prostate cancer (mCRPC) and localized high-risk prostate cancer with PI3K pathway activation.
