iPD1 CD19 eCAR T cells is an autologous, fourth-generation ("armored") chimeric antigen receptor (CAR) T-cell therapy developed by Peking University for the treatment of relapsed or refractory B-cell lymphoma. The therapy consists of patient-derived T cells genetically engineered using a lentiviral vector to express a second-generation CAR targeting the CD19 antigen (utilizing a 4-1BB costimulatory domain) alongside a short hairpin RNA (shRNA) cassette designed to knockdown the expression of Programmed Cell Death Protein 1 (PD1). By incorporating intrinsic PD1 inhibition, the therapy aims to prevent the induction of the PD1/PD-L1 immunosuppressive pathway upon T-cell activation, thereby enhancing the persistence and anti-tumor activity of the CAR T cells within the tumor microenvironment. It is currently being evaluated in Phase 1 clinical trials.
