IPH2101 + lenalidomide is a combination therapy under investigation for the treatment of multiple myeloma. IPH2101 (also known as 1-7F9) is a monoclonal antibody that targets and blocks inhibitory killer cell immunoglobulin-like receptors (KIRs) on natural killer (NK) cells, thereby preventing negative regulatory signals and enhancing NK-cell-mediated cytotoxicity against multiple myeloma cells. Lenalidomide is an immunomodulatory drug that augments NK-cell function, upregulates ligands for activating NK-cell receptors on tumor cells, and exerts antineoplastic, antiangiogenic, and immune-stimulatory effects. The combination aims to synergistically enhance the immune response against multiple myeloma by both blocking inhibitory signals to NK cells (via IPH2101) and boosting their activity (via lenalidomide). This regimen has been evaluated in phase 2 clinical trials for patients with multiple myeloma[1][2][4][7].
