Ipilimumab + AS01B is an investigational immunotherapy combination being studied in clinical trials for cancer treatment. Ipilimumab is a monoclonal antibody that blocks cytotoxic T-lymphocyte associated protein-4 (CTLA-4), enhancing T-cell activation and proliferation. AS01B is a liposome-based adjuvant containing two immunostimulants: monophosphoryl lipid-A and QS21 (a saponin isolated from the Quillaja saponaria tree). AS01B induces strong recruitment of conventional dendritic cells (cDC1 and cDC2) and enhances antigen presentation to both CD4+ and CD8+ T cells. The combination is being investigated particularly for intratumoral administration in melanoma and other cancers, often alongside other immunotherapies like nivolumab.
