Overview

Ipilimumab + gemcitabine is a combination therapy investigated for the treatment of advanced pancreatic ductal adenocarcinoma (PDAC). Ipilimumab is a fully human monoclonal antibody that targets cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), an immune checkpoint molecule that downregulates T-cell activation. By blocking CTLA-4, ipilimumab enhances T-cell-mediated anti-tumor immune responses. Gemcitabine is a nucleoside analog chemotherapy that inhibits DNA synthesis. This specific combination was evaluated in a Phase Ib dose-escalation study (NCT01473940) led by Northwestern University to determine safety and efficacy in patients with unresectable or recurrent pancreatic cancer.
