**Ipilimumab + gp100 peptide vaccine** is a combination therapy studied primarily for advanced melanoma. Ipilimumab is a fully human monoclonal antibody that targets cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), acting as an immune checkpoint inhibitor to promote T cell-mediated anti-tumor responses. The gp100 peptide vaccine consists of synthetic peptides that correspond to sequences from the melanoma antigen glycoprotein 100 (gp100), designed to elicit tumor-specific immune responses by stimulating T cells specific for gp100-presenting tumor cells. The combination does not appear to provide additional survival benefit beyond ipilimumab alone, but both approaches are superior to gp100 vaccine alone in metastatic melanoma[1][2][5][6][8]. The combination, as studied, was restricted to HLA-A2-positive patients.
