A combination immunotherapy regimen, also known as IPI-Biotherapy, developed and investigated by the M.D. Anderson Cancer Center for the treatment of metastatic melanoma. The regimen consists of the monoclonal antibody ipilimumab (a CTLA-4 checkpoint inhibitor), the type I interferon cytokine interferon alfa-2b, and the recombinant interleukin-2 cytokine aldesleukin. This triple combination was designed to synergistically enhance the anti-tumor immune response by removing inhibitory signals on T-cells (via ipilimumab) and providing direct stimulatory signals to promote the activation and expansion of cytotoxic T-lymphocytes and natural killer cells (via interferon and aldesleukin). It was primarily evaluated in a Phase I/II clinical trial (NCT00494338) for patients who had previously failed cytotoxic chemotherapy, although the study was ultimately terminated due to slow patient accrual.
