Overview

Ipilimumab + pexmetinib is an investigational combination therapy for advanced solid tumors, including melanoma, kidney cancer, and non-small cell lung cancer. Ipilimumab is a monoclonal antibody that acts as a checkpoint inhibitor targeting cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), thereby enhancing immune responses against tumor cells. Pexmetinib (ARRY-614) is an orally bioavailable small-molecule inhibitor of p38 MAP kinase and Tie2 kinase with antineoplastic, anti-inflammatory, and antiangiogenic activities. By inhibiting these kinases, pexmetinib may reduce pro-inflammatory cytokine production and tumor angiogenesis while promoting tumor cell death. The combination aims to leverage both immune activation via CTLA-4 blockade and direct inhibition of tumor-promoting pathways through kinase inhibition[1][5][7][9].
