iprindole + lithium carbonate

This is a combination therapy consisting of iprindole, a tricyclic antidepressant, and lithium carbonate, a mood stabilizer. Iprindole is an atypical tricyclic antidepressant that does not inhibit monoamine reuptake but may sensitize forebrain neurons to serotonin (5-HT). Lithium carbonate is primarily used as a mood stabilizer in bipolar disorder and works by enhancing serotonergic neurotransmission among other mechanisms. The combination has been studied for use in patients with major unipolar depression who are refractory to tricyclic antidepressants alone. Clinical studies have shown that adding lithium carbonate to ongoing iprindole treatment can result in rapid and significant improvement of depressive symptoms within 48 hours, likely due to enhanced serotonergic activity on sensitized neurons[1][4][3]. This approach is sometimes referred to as "lithium augmentation" of antidepressant therapy.
