Ir(ppy)2(pytr-Sq-girentuximab) is an experimental antibody-drug conjugate (ADC) designed for the dual purposes of photodynamic detection (PDD) and photodynamic therapy (PDT). It consists of a phosphorescent organometallic iridium(III) complex, [Ir(ppy)2(pytr-Sq)], covalently linked to the monoclonal antibody girentuximab via a squaramide-polyethylene glycol (PEG) linker. Girentuximab specifically targets **carbonic anhydrase IX (CAIX)**, a cell-surface enzyme that is highly overexpressed in hypoxic tumor environments, including clear cell renal cell carcinoma (ccRCC), glioblastoma, and triple-negative breast cancer. The iridium(III) complex serves as both an emissive probe for fluorescence-guided surgical resection and a photosensitizer that, upon irradiation with visible light, generates reactive oxygen species (ROS) to induce localized cytotoxicity. This conjugate represents a targeted approach to treating and visualizing CAIX-positive solid tumors.
