Overview

Iratumumab (MDX-060) is a fully human IgG1 monoclonal antibody directed against CD30 (TNFRSF8), a cell surface antigen highly expressed on Reed-Sternberg cells in Hodgkin lymphoma and neoplastic cells in anaplastic large cell lymphoma (ALCL). Developed by Medarex in collaboration with Bristol Myers Squibb, iratumumab was designed to induce antitumor activity through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and signal transduction inhibition. While it demonstrated a manageable safety profile in Phase I/II clinical trials for relapsed or refractory CD30-positive lymphomas, its clinical development was ultimately discontinued as it did not show sufficient monotherapy efficacy to compete with emerging therapies like the antibody-drug conjugate brentuximab vedotin.
