irinotecan + cetuximab + envafolimab

This is a combination therapy consisting of three agents: irinotecan, cetuximab, and envafolimab. Irinotecan is a topoisomerase I inhibitor that interferes with DNA replication in cancer cells, leading to cell death. Cetuximab is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking its activation and downstream signaling pathways involved in tumor cell proliferation and survival. Envafolimab is a humanized single-domain anti-PD-L1 antibody administered subcutaneously; it blocks the interaction between PD-L1 on tumor cells and PD-1 on T-cells, thereby enhancing anti-tumor immune responses. This triple regimen is being investigated as a rechallenge treatment for patients with RAS/BRAF wild-type metastatic colorectal cancer who have previously benefited from irinotecan and cetuximab[1][2][6].
