irinotecan + cisplatin + simvastatin

Irinotecan + cisplatin + simvastatin is a combination chemotherapy regimen investigated by the National Cancer Center, Korea, for the treatment of extensive-disease small cell lung cancer (ED-SCLC). This regimen combines two standard cytotoxic agents—irinotecan (a topoisomerase I inhibitor) and cisplatin (a platinum-based DNA cross-linking agent)—with simvastatin, an HMG-CoA reductase inhibitor. The addition of simvastatin was based on preclinical evidence suggesting that statins may enhance the efficacy of chemotherapy or overcome resistance in lung cancer cells. The combination was evaluated in a randomized Phase II trial (NCT00931515) to determine if adding simvastatin to the standard irinotecan/cisplatin (IP) doublet improved clinical outcomes in chemo-naive patients.
