irinotecan + raltitrexed

Irinotecan + raltitrexed is a combination chemotherapy regimen consisting of two small molecule drugs, irinotecan and raltitrexed. Irinotecan is a topoisomerase I inhibitor that interferes with DNA replication in cancer cells, leading to cell death. Raltitrexed is a thymidylate synthase inhibitor that disrupts DNA synthesis by inhibiting the formation of thymidine, an essential nucleotide for DNA replication. This combination has been primarily studied and used in the treatment of advanced or metastatic colorectal cancer (CRC), especially as second-line therapy after failure of first-line regimens such as oxaliplatin and fluorouracil. It has also been investigated for use in esophageal squamous cell carcinoma as salvage chemotherapy. The regimen is sometimes referred to by the acronym TOMIRI.
