This is a combination therapy consisting of two agents: irinotecan liposome (II), a nanoparticle formulation of the topoisomerase I inhibitor irinotecan, and ivonescimab, an investigational bispecific monoclonal antibody targeting both programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A). Irinotecan liposome delivers cytotoxic chemotherapy directly to tumor cells by inhibiting DNA replication through topoisomerase I inhibition. Ivonescimab simultaneously blocks PD-1 to enhance immune-mediated tumor cell killing and inhibits VEGF-A to disrupt tumor angiogenesis. This combination is being investigated as a second-line treatment for small cell lung cancer in patients who have failed first-line platinum-based chemotherapy with or without checkpoint inhibitors[1][2][6][7][9].
