Overview
Irinotecan liposome (COF007) is a liposomal formulation of the topoisomerase I inhibitor irinotecan, being developed by Lee's Pharmaceutical through its subsidiary China Oncology Focus Limited. Irinotecan is a semi-synthetic derivative of camptothecin and acts as a prodrug that is metabolically converted to its active metabolite, SN-38. SN-38 binds to the DNA topoisomerase I enzyme, stabilizing the cleavable complex and preventing the religation of single-strand DNA breaks. This leads to lethal double-strand DNA breaks during replication and subsequent cell death. The liposomal delivery system is designed to improve the pharmacokinetic profile of irinotecan by increasing the duration of exposure to SN-38 within the tumor microenvironment while potentially reducing systemic toxicity. COF007 is primarily being investigated for the treatment of advanced solid tumors, with a focus on pancreatic cancer.
