Drug intelligence / Profile preview
Irinotecan liposome is a liposomal-encapsulated formulation of irinotecan hydrochloride developed to overcome the pharmacological and clinical limitations of conventional irinotecan. The active substance irinotecan is a topoisomerase I inhibitor that blocks the enzyme topoisomerase I, which is involved in DNA replication. By blocking this enzyme, cancer cells are prevented from multiplying and eventually die. In this formulation, irinotecan is enclosed in pegylated liposomes (tiny fat particles coated with polyethylene glycol), which are designed to accumulate in tumors and release the drug slowly over time. This extends circulation time, reduces premature metabolism, and allows sustained drug action at tumor sites while decreasing systemic toxicity. The formulation was initially developed by Merrimack Pharmaceuticals and is now manufactured by Ipsen Biopharmaceuticals. It is primarily indicated for the treatment of metastatic pancreatic adenocarcinoma.
