irinotecan liposome + S-1 is a combination chemotherapy regimen being evaluated for the treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC). The regimen consists of irinotecan liposome, a nanoparticle-encapsulated formulation of the topoisomerase I inhibitor irinotecan, and S-1, an oral fluoropyrimidine combination drug containing tegafur (a 5-fluorouracil prodrug), gimeracil, and oteracil. This combination is specifically being studied by Sun Yat-sen University in patients whose disease has progressed after treatment with PD-1 or PD-L1 inhibitors. Irinotecan liposome is designed to enhance the delivery of the active metabolite SN-38 to tumor tissues while reducing systemic toxicity, and S-1 provides synergistic anti-tumor activity through the inhibition of thymidylate synthase.
