Irofulven + capecitabine is a combination of two small molecule chemotherapeutic agents used in clinical trials for the treatment of advanced solid tumors and certain cancers such as thyroid cancer. Irofulven is a semisynthetic derivative of illudin S that acts as an alkylating agent causing DNA damage and apoptosis in tumor cells. Capecitabine is an oral prodrug that is enzymatically converted to 5-fluorouracil (5-FU) within tumor tissue; 5-FU inhibits thymidylate synthase and interferes with DNA synthesis, leading to cell death. The combination has been studied for synergistic antitumor activity[2][4][5][6].
