irpagratinib + ABSK043 is a combination therapy being investigated for the treatment of unresectable or metastatic hepatocellular carcinoma (HCC) with FGF19 overexpression. The combination consists of irpagratinib (ABSK-011), an oral, highly selective, and irreversible small molecule inhibitor of fibroblast growth factor receptor 4 (FGFR4), and ABSK043, an oral small molecule inhibitor of the programmed death-ligand 1 (PD-L1). Irpagratinib targets the FGF19-FGFR4 signaling axis, a known driver of tumor growth in a subset of HCC patients, while ABSK043 acts as an immune checkpoint inhibitor to restore the anti-tumor activity of T-cells. This combination approach aims to provide synergistic efficacy by simultaneously inhibiting oncogenic signaling and enhancing the immune response against the tumor.
