Overview

ABSK-011 + toripalimab + bevacizumab is an investigational triplet combination regimen being developed by Abbisko Therapeutics for the first-line treatment of advanced or unresectable hepatocellular carcinoma (HCC), specifically in patients with FGF19 overexpression. The regimen consists of irpagratinib (ABSK-011), a highly selective oral small-molecule inhibitor of Fibroblast Growth Factor Receptor 4 (FGFR4); toripalimab, a humanized monoclonal antibody targeting the Programmed Cell Death Protein 1 (PD-1) receptor; and bevacizumab, a monoclonal antibody that inhibits Vascular Endothelial Growth Factor (VEGF). By targeting three distinct pathways—FGFR4 signaling, immune evasion, and angiogenesis—the combination seeks to provide a synergistic therapeutic effect in HCC patients whose tumors are driven by FGF19/FGFR4 alterations.
