Overview

IRX4204 + erlotinib is a combination therapy under clinical investigation, primarily for advanced non-small cell lung cancer (NSCLC). IRX4204 is a highly selective, potent agonist of Retinoid X Receptor (RXR), a nuclear receptor involved in regulating gene expression. Erlotinib is a well-established epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. The combination aims to inhibit multiple cancer cell growth pathways synergistically. Preclinical data suggest the combination may overcome resistance mechanisms and provide additive anti-proliferative effects, affecting signaling pathways including Src, PI3K/AKT, ribosomal S6, AMPK, TGF-β, and angiogenesis. IRX4204 is developed by IO Therapeutics and is orally administered, as is erlotinib.
