Isatuximab + belantamab mafodotin + dexamethasone is a combination regimen evaluated for the treatment of relapsed or refractory multiple myeloma (RRMM). Isatuximab is a monoclonal antibody targeting CD38, leading to antitumor activity through immune-mediated mechanisms. Belantamab mafodotin is an antibody-drug conjugate targeting B-cell maturation antigen (BCMA); it delivers a cytotoxic agent (MMAF) to multiple myeloma cells, resulting in apoptosis. Dexamethasone is a corticosteroid used to augment anti-myeloma effects and reduce inflammation. This triplet aims to exploit complementary mechanisms—immune modulation, targeted cytotoxin delivery, and anti-inflammatory/pro-apoptotic effects—to improve efficacy in refractory or heavily pretreated myeloma patients[7][1].
