Overview

This drug is a combination of three agents: **isatuximab** (a monoclonal antibody targeting CD38), **evorpacept** (an Fc fusion protein acting as a CD47-blocking myeloid checkpoint inhibitor), and **dexamethasone** (a synthetic glucocorticoid corticosteroid). The combination is being investigated in relapsed/refractory multiple myeloma (RRMM). Isatuximab promotes immune-mediated killing of plasma cells by targeting CD38, a molecule highly expressed on malignant plasma cells. Evorpacept inhibits the "don't eat me" signal by blocking CD47, thereby enhancing phagocytosis of cancer cells by macrophages. Dexamethasone reduces inflammation and also has cytotoxic effects on plasma cells. All three are being tested together in clinical trials for synergistic anti-myeloma activity, especially in immunotherapy-resistant settings[4][5].
