ISIS 113715 + sulfonylurea is a combination therapy of a second-generation antisense oligonucleotide drug (ISIS 113715) with an oral sulfonylurea agent, studied for the treatment of type 2 diabetes mellitus in patients with inadequate glycemic control on sulfonylurea monotherapy. ISIS 113715 specifically targets and inhibits protein tyrosine phosphatase 1B (PTP-1B), a negative regulator of insulin receptor signaling, thereby enhancing insulin sensitivity. Sulfonylureas act by stimulating pancreatic beta-cell insulin secretion through inhibition of ATP-sensitive potassium channels. The combination aims to complement the insulin-sensitizing effect of ISIS 113715 with the insulinotropic effect of sulfonylureas. Clinical studies demonstrated improved glycemic parameters and lipid profiles in type 2 diabetes patients on stable sulfonylurea doses with the addition of ISIS 113715. Developed by Isis Pharmaceuticals, this combination has been investigated in phase 2 clinical trials[1][3][5][7].
