A combination immunotherapy drug candidate comprised of two tumor-associated antigens: gp100 (glycoprotein 100) and TRP-2 (tyrosinase-related protein 2), designed to activate the immune system against tumor cells expressing both antigens. This combination generally takes the form of a DNA vaccine or engineered immunotherapy platform, with the most studied example being SCIB1 ImmunoBody, which includes epitopes from both gp100 and TRP-2 to induce robust CD8+ and CD4+ T cell responses. Commonly developed and studied in preclinical and clinical settings for melanoma and glioblastoma multiforme (GBM), its mechanism is to prime and expand T-cell populations capable of recognizing and killing tumor cells expressing these melanoma-associated antigens. The approach is often tested in combination with immune checkpoint blockade (such as anti-PD-1 therapies) to enhance antitumor immunity, particularly in tumors with high PD-L1 expression, such as GBM.
