Overview

Isoniazid + rifampicin is a fixed-dose combination of two antibiotics used primarily to treat tuberculosis (TB). Isoniazid inhibits the synthesis of mycolic acids, essential components of the mycobacterial cell wall, by targeting enoyl-acyl carrier protein reductase after activation by bacterial catalase-peroxidase. Rifampicin inhibits bacterial DNA-dependent RNA polymerase, thereby suppressing RNA synthesis and subsequent protein production. This combination is typically administered orally and may be used with other antituberculosis medications. It appears on the World Health Organization's List of Essential Medicines due to its importance in TB treatment[1][2][5][6].
