Kinisoquin + Zafirlukast is an oral small-molecule combination therapy being developed by Quercis Pharma AG. The combination pairs isoquercetin (Kinisoquin), a Protein Disulfide Isomerase (PDI) inhibitor with antithrombotic properties, with zafirlukast (Accolate), a leukotriene receptor antagonist. The two agents are believed to act synergistically by targeting PD-L1 and VEGF pathways, inhibiting PDI, and modulating Interleukin-17 (IL-17) signaling. These pathways are implicated in thrombosis, cancer progression, and neurological diseases. The combination is described as Phase 2-ready and is being investigated for the treatment of various cancers and neurological disorders, with reported potential synergy when used alongside checkpoint inhibitors like pembrolizumab or standard chemotherapy regimens such as cisplatin plus gemcitabine. Preclinical data in ovarian cancer models have demonstrated cooperative suppression of tumor growth and thromboinflammatory signaling.
