R.I.T.E. is a combination chemo-immunotherapy regimen consisting of isotretinoin (13-cis retinoic acid), recombinant interferon alfa-2b, docetaxel (Taxotere), and estramustine phosphate sodium. Developed and studied by the University of Medicine and Dentistry of New Jersey (UMDNJ), this regimen was designed to treat hormone-refractory metastatic prostate cancer. The combination leverages multiple mechanisms: docetaxel stabilizes microtubules to inhibit cell division, estramustine disrupts microtubules and provides estrogenic effects, interferon alfa-2b acts as an immunomodulator and anti-proliferative agent, and isotretinoin promotes cell differentiation. By combining these diverse modalities, the R.I.T.E. regimen aims to overcome drug resistance and enhance tumor cell death in advanced prostate cancer patients.
