Overview

**Ispinesib + carboplatin** is a combination of two anti-cancer agents: ispinesib, a **small molecule inhibitor of kinesin spindle protein (KSP)**, and carboplatin, a platinum-based cytotoxic chemotherapy. Ispinesib inhibits mitosis by blocking KSP, a motor protein essential for bipolar mitotic spindle formation and proper cell cycle progression, forcing cells to arrest during mitosis and undergo apoptosis. Carboplatin induces DNA cross-linking and damage, inhibiting DNA synthesis and function, leading to cell death. This combination has been studied in early-phase clinical trials to evaluate safety, tolerability, dosing, and antitumor activity, especially in patients with advanced or metastatic solid tumors including breast cancer[1][2]. Ispinesib was originally developed by Cytokinetics in collaboration with GlaxoSmithKline (GSK). Clinical trials have demonstrated that the combination is generally tolerable, with dose-limiting toxicities consistent with those of the individual agents; grade 4 neutropenia is the main limiting toxicity[2]. The combination is primarily under investigation for oncology indications and has not been approved for clinical use.
