Overview

This is a combination regimen consisting of three agents: - **Ivonescimab (AK112):** A bispecific monoclonal antibody targeting both PD-1 and VEGF pathways. It acts as an immune checkpoint inhibitor and anti-angiogenic agent. Ivonescimab is under investigation for its antitumor activity in various cancers. - **Albumin-bound paclitaxel (nab-paclitaxel):** A nanoparticle formulation of the microtubule inhibitor paclitaxel bound to albumin to enhance delivery and reduce toxicity compared to conventional formulations. - **Cisplatin:** A platinum-based chemotherapeutic that induces DNA crosslinking and apoptosis in rapidly dividing cells. The combination is being studied as a first-line treatment for advanced malignancies such as esophageal cancer[1][2]. The rationale is to combine immunotherapy (ivonescimab) with cytotoxic chemotherapy (albumin-bound paclitaxel and cisplatin) for synergistic antitumor effects.
