Overview

This combination therapy consists of ivonescimab (AK112), a first-in-class bispecific antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), administered alongside the CapeOX chemotherapy regimen (capecitabine and oxaliplatin) and radiotherapy. It is currently being evaluated in a Phase II clinical trial (NCT05795387) sponsored by Fudan University for the treatment of patients with unresectable metastatic microsatellite stable (MSS) colorectal cancer. The regimen aims to leverage the synergistic effects of dual checkpoint and angiogenesis inhibition with cytotoxic chemotherapy, potentially enhanced by the abscopal effects of radiotherapy, to overcome the typically immunosuppressive microenvironment of MSS colorectal tumors.
