Overview

Ivonescimab + gumagalimab is a combination of two monoclonal antibodies developed for cancer immunotherapy. Ivonescimab is a bispecific antibody targeting PD-1 (Programmed cell death protein 1) and VEGF (Vascular endothelial growth factor), designed to inhibit both immune checkpoint activity and tumor angiogenesis. This dual targeting aims to recalibrate the tumor microenvironment to favor anti-tumor immunity and inhibit blood vessel formation, improving efficacy in lung cancer and potentially overcoming immune resistance[1][5]. Gumagalimab (also known as AK119) is a monoclonal antibody targeting TIM-3 (T cell immunoglobulin and mucin-domain containing-3), an immune checkpoint receptor expressed on T cells and myeloid cells. TIM-3 blockade aims to further enhance immune response in the tumor microenvironment. This combination is being investigated in clinical trials for advanced and metastatic non-small cell lung cancer (NSCLC), including EGFR-mutant and squamous/non-squamous histologies.
