Overview

Ivonescimab + ipilimumab is an investigational combination immunotherapy regimen consisting of ivonescimab (SMT112/AK112), a novel bispecific antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), and ipilimumab (Yervoy), a recombinant human monoclonal antibody targeting cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4). This combination is designed to simultaneously block the PD-1/PD-L1 pathway, inhibit VEGF-mediated angiogenesis, and promote T-cell activation via CTLA-4 inhibition. The regimen is being evaluated in a Phase II clinical study (SMT112-216 / AK112-216 / NCT07877194) for the first-line treatment of patients with advanced clear cell renal cell carcinoma (ccRCC).
