Ivonescimab (AK112) is a first-in-class humanized IgG1 bispecific antibody that simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF). By dual-targeting these pathways, it aims to enhance anti-tumor activity through both immune checkpoint inhibition and anti-angiogenesis. This specific combination involves ivonescimab administered alongside platinum-based chemotherapy (such as cisplatin or carboplatin) for the treatment of advanced non-small cell lung cancer (NSCLC). It is particularly being investigated in patients with driver mutations (EGFR, ALK, or ROS1) who have progressed after initial targeted therapies, as well as in first-line settings for patients without driver mutations.
