Overview

Ivonescimab + sacituzumab govitecan is an investigational combination therapy being evaluated for the treatment of triple-negative breast cancer (TNBC), specifically in patients with brain metastases. Ivonescimab (AK112) is a first-in-class bispecific antibody that simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), designed to combine immunotherapy with anti-angiogenesis. Sacituzumab govitecan is a TROP2-directed antibody-drug conjugate (ADC) consisting of a humanized monoclonal antibody conjugated to the topoisomerase I inhibitor SN-38. This combination aims to leverage the synergistic effects of immune checkpoint inhibition, anti-angiogenesis, and cytotoxic chemotherapy delivered via a targeted ADC to overcome resistance in advanced malignancies.
