Overview

ivonescimab + VG2025 is an investigational combination immunotherapy regimen being developed for the treatment of advanced colorectal cancer with liver metastases (CRLM). The regimen consists of ivonescimab (AK112), a first-in-class bispecific antibody that simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), and VG2025, a next-generation oncolytic herpes simplex virus type 1 (HSV-1). This combination therapy is designed to leverage multiple synergistic mechanisms: VG2025 induces precise oncolysis and cross-primes tumor antigens to activate tumor-specific T-cells, while ivonescimab provides dual checkpoint blockade and anti-angiogenic effects to overcome the immunosuppressive tumor microenvironment. The therapy is currently being evaluated in Phase Ib/II clinical trials for patients who have progressed on standard therapies.
