This is a combination of four distinct anticancer drugs, each with different mechanisms of action. Ivosidenib is an IDH1 inhibitor used for IDH1-mutated myeloid malignancies. Venetoclax is a BCL-2 inhibitor that promotes apoptosis in cancer cells. Gilteritinib is a FLT3 inhibitor used in FLT3-mutated acute myeloid leukemia. Selinexor is an XPO1 inhibitor used in multiple myeloma and diffuse large B-cell lymphoma. While ivosidenib + venetoclax (with or without azacitidine) has been studied in clinical trials for IDH1-mutated myeloid malignancies, and selinexor has shown synergy with FLT3 inhibitors like gilteritinib, there is no evidence of all four drugs being used together as a single therapeutic regimen.
