Ixabepilone + trastuzumab is a combination therapy used primarily in the treatment of HER2-positive metastatic or locally advanced breast cancer. Ixabepilone is a semisynthetic analog of epothilone B and acts as a microtubule stabilizer, disrupting cell division by binding to β-tubulin and promoting microtubule polymerization, which leads to apoptosis. Trastuzumab is a monoclonal antibody that targets the human epidermal growth factor receptor 2 (HER2), inhibiting downstream signaling pathways involved in cell proliferation and survival. The combination has shown efficacy in patients with HER2-positive breast cancer, including those previously treated with taxanes or anthracyclines, and can be used as first-line or subsequent therapy for metastatic disease[1][4][9]. Clinical studies have demonstrated encouraging response rates and manageable toxicity profiles for this regimen.
