This is a combination therapy consisting of **ixazomib**, an oral proteasome inhibitor, plus one or more **chemotherapy** agents. The primary and FDA/EMA-approved chemotherapeutic combination is **ixazomib with lenalidomide and dexamethasone** for the treatment of patients with relapsed or refractory multiple myeloma. **Ixazomib** is a reversible inhibitor of the chymotrypsin-like activity of the **protein proteasome subunit beta type-5 (PSMB5)**, disrupting protein homeostasis and inducing apoptosis in cancer cells. **Lenalidomide** (typically considered an immunomodulatory drug, sometimes classified as chemotherapy in clinical protocols) works by affecting cellular signaling and the immune microenvironment, while **dexamethasone** is a corticosteroid that enhances antimyeloma effects. Other cytotoxic chemotherapeutic agents are sometimes used with ixazomib in clinical studies, but “ixazomib + chemotherapy” commonly refers to this triple regimen. Ixazomib is developed and marketed by **Takeda** under the brand name Ninlaro and is the first oral proteasome inhibitor approved for multiple myeloma therapy[1][5][7][8][9].
