This is a combination regimen consisting of **ixazomib**, an oral small molecule proteasome inhibitor; **pegylated liposomal doxorubicin** (PLD), a cytotoxic anthracycline antibiotic encapsulated in a liposomal carrier to reduce cardiotoxicity; and **dexamethasone**, a synthetic glucocorticoid corticosteroid. Ixazomib inhibits the chymotrypsin-like activity of the 20S proteasome, blocking protein degradation and inducing apoptosis in myeloma cells[5][1]. Pegylated liposomal doxorubicin intercalates DNA, inhibits topoisomerase II, and generates cytotoxic free radicals. Dexamethasone has anti-inflammatory, antiemetic, immunosuppressive, and direct antitumor activity in certain hematologic malignancies, acting via the glucocorticoid receptor[6]. This triple drug regimen is primarily used in the treatment of **relapsed or refractory multiple myeloma** and is being investigated in clinical trials for this indication. The combination leverages the targeted cytotoxic effects of ixazomib and doxorubicin with the immunomodulatory and supportive effects of dexamethasone.
