The combination of ixazomib, pomalidomide, and dexamethasone is an all-oral regimen used primarily for the treatment of relapsed or refractory multiple myeloma. Ixazomib is a proteasome inhibitor that disrupts protein degradation in cancer cells leading to cell death. Pomalidomide is an immunomodulatory agent that enhances immune response against myeloma cells and inhibits their growth. Dexamethasone is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressive properties; it also induces apoptosis in certain cancer cells. This triplet therapy has demonstrated improved progression-free survival and overall response rates compared to doublet regimens in lenalidomide-refractory multiple myeloma patients[2][5][8]. The regimen leverages synergistic mechanisms to overcome resistance seen with prior therapies.
